Hi all
I would like to know the appropriate pore size culture inserts to be used for skin organotypic cultures. In many papers they mention the use of 6 well inserts with 3um pore size while few papers use 24 well inserts of 0.4um pore size.
How does pore size affect organotypic culture?My culture are keratinocyte cells grown over collagen and fibroblasts, then allowed air liquid interface for differentiation.
I would really appreciate any suggestions/recommendations.

for organotypics you want a good mass transfer from the culture medium so I woudl go for the bigger pore sizes, at least 3 microns , I dont see the point in 0.4 um. ,, please see above guide, best wishes, Michael
I agree with Michael. However both will work for your application. 0.4 um is more than large enough for water molecules. Just don't go too big, else your cells may migrate through the semi-permeable barrier!
Why do people use different sizes? Probably because that's what they had on hand! No better reason.
