My iPSC clones in mTeSR plus culture had bad performance recently. There seem to be excessive dead cells and clumps sticking to the clones. The dead cells can not be removed by flapping the dish. This situation lasted for a week. The newly thawed one also had a similar problem. The overall morphology of clones looks good and there seem to be no signs of bacteria contamination. My colleagues had two explanations: one is the contamination of uncertain pathogen, and the "dead cells or clumps" are actually the pathogen. The pathogen may not be so lethal that the overall growth of the clone is good. The other is the spontaneous differentiation caused by overcrowded clones before the passaging since the clones grow to about 80-90% crowd before the passaging and the general status did not look well. So, I decided to discard these clones but I really want to know the reason why it looks bad.
