We prepared PLGA np of different sizes ranging from 80, 100, 120,...., 200, 220nm. Do you think it has an effect on cell lines. Cellular uptake behaviour internalization and MTT assay.??
Why not prepare NP’s containing FITC for each size and set up an assay to measure uptake?
Effect could be different, as smaller NPs in most cases penetrate better and faster into cells. Dr. William Richard Webb gave a very good suggestion, you could track your NPs and see what happens. Of course, you also should perform MTT assay in parallel using different size NPs as they all could be non toxic (toxicity does not have to be correlated directly with cell penetration).

I performed MTT assay as well Coumarin6 fluorescenct dyes labelled cellular internalization study on different mammalian cell lines. But I am unable to differentiate their effects. There should be differences in uptake and cytotoxity rate. As size must show some sort of differences. How I can report the size affects these results??
