Somebody please suggest me about the growth media for EPC primary culture. A number of growth factors present in EGM-2 of LONZA but in some of the previous work only VEGF and FBS has been used as growth supplement for the same along with basal media and they have done characterization and all functional studies within 7-10 days of culture. So endothelial basal media (EBM) or M199 with VEGF and 20% FBS/FCS only would be sufficient or not for normal growth and proliferation of EPC.
When I did VEGF activation assays I used basal media [Lonza] and added VEGF of known concentrations and I think 0.1% FBS. But my experiments ran usually not longer than 24h.
20% FBS??? That sounds a hell of a lot to me. You surly mean 2%. You can grow endothelial cells in 2%. 2% is just about at the starvation limit. They will grow slowly but will be fine.
