I am facing a problem in maintaining the cell line of HepG2 and HepG2.2.15. I have used plasmocin Tm treatment (Invivo gen) Cat.#ant-mpt Lot #MPT-41-01 for more than a month. I have also used gentamicin, pepzo antibiotics. But the problem persists. Any suggestion is highly appreciated.
agree with Malcolm. Mycoplasma contamination can be very stubborn. It is best that you discard your current lot, sterilize and decontaminate everything thoroughly, and then start afresh.

Plasmocin is a good broad-spectrum ant-mycoplasma agent. Usually the contamination comes under control in a month's time. But this is not always possible.
Having mycoplasma contamination affect cells in culture because it can bring about changes in the receptors present on the cell surface. So if you use these cells for any assays the results would not be as per expectations.
I suggest you don't waste time in decontaminating. Instead, use fresh cells and restart because it seems that the contamination is of a serious type. It is always better to find and then treat the source of contamination.
