I have also used DNS assay to calculate the activity.

This is the method I used to calculate the activity

If the OD 540 value is 1.04 OD,    then it is equal to 0.53mg of D-galacturonic acid( from the standard graph of D-galacturonic acid (plot wit d D-galacturonic acid concentration of 1mg/ml)

Details of my DNS assay are;

Vol of substrate (1% citrus pectin) = 0.5 ml

vol of crude enzyme               = 0.5 ml

vol of DNS reagent                 = 1.5 ml

Dilution factor                          = 10

Reaction time                          = 15 min

Then what will be the enzyme activity? Please help me to calculate the enzyme activitu (U/mi)?
