Hello all! I plated some secretory tissue from the udder of a dairy cow. I am trying to isolate mammary epithelial cells. I found these cells on one of my plates, are these developing mammary epithelial cells or are they some type of connective tissue? They have a cobblestone morphology, but not completely sure what they are because they look slightly different than what I'm used to.
The squamous appearance imply that these are probably epithelial cells. However, the morphology of cells is not enough to characterize them. I recommend using flowcytometry technique with specific antibodies to characterize your cells precisely.
Thank you Arman. Since you are here, I would also like to ask if you have experience coating plates with collagen or if you buy pre-coated plates. If so, how do I decide on the correct collagen for my tissues to attach. Since I am using bovine tissue, is it best to use bovine collagen? Or does it not make a difference.

Unfortunately, I have no experience on using collagen-coated plates. Could you please tell me why are you going to use collagen-coated plates? If you want to increase the adhesiveness of your cells, I propose simply using gelatin for coating your plates. It usually works well for primary cultures.
