Dear Mahmoud Kiani what exactely do you mean?? Are you looking for a method to identify the phenols in your extracts?? In this case you should use methods such as HPLC-DAD, LC-MS and so on.

On the other hand, if you are wondering if your extraction works well (and thus the phenolic compounds are present in your extract) you can check it by performing other extractions from the same oil, determining the total phenolic content. In this way you can be sure that in one step you are able to extract all or you need to change method or perform more then one step of extraction.

Yes... I mean all the phenolic costituents are from the plant, so you can find some of them in the essential oil instead of others, depending on the solubility etc. So, if you start your extraction from the plant material, I think you can expect to extract even those phenolics you find in the essential oil and probably even others.

Yes of course if you use a solvent such as ethanol or methanol to extract phenolic compounds, the solvent will contain them. Different phenolics have different "extractability", but I would expect that all the phenolic compounds that are easily extractable (in contrast, for example, to the fraction of bound phenolics) will be found in the solvent.

Whilst you will probably analyse just the total phenolic content and therefore you won't be targeting specific phenolic compounds, you will found a wide range of phenolic compounds including the ones you mentioned. You have different extractions if you use ethanol or hexanal, particuarly in terms of lipophilic and hydrophilic substances.

in your solvent extraction you'll get all compounds either phenols, terpenes,etc. If you are searching pulegone, the major compound of Mentha pulegium or menthol from Mentha piperita. I think it's better to get these oxygenated monoterpenes by extraction of essentail oils directly

You are right, and I have the essential oil content of my samples too. But the primary objective of my work is determination of scolocidal activity of crude extract from Mentha sp. I want to make sure phenolic terpenoids are also present in the extract.
