This is my first time using commercialised PCI (25:24:1) solution. After adding equal amount of PCI into my supernatant, two of my samples did not form two layers as they should be. After centrifuging at 13 000g for 10 minutes, one showed a coagulation at the bottom of the tube whereas the other one, remains unchanged. I centrifuged the latter one to at least precipitate it but it just wont. This has never happened before.

Another unrelated situation, what would happened if an oxidised phenol is used in dna extraction?

Do you notice two phases in your PCI? You should use the lower phase as the upper phase only contains water and buffer.
Chun Keat Tan No, i didnt notice as the PCI was in a dark brown glass bottle. Do you think this affected the result? Let say if i did used the water phase, why are there two different results? one that coagulates and one that doesnt?
My PCI is in amber bottle so I still can see the two phases with light background. May I know what are your samples?
