Hi I am working on peptides , I have synthesised the Cyclic peptide , i want to purify I am running the HPLC of this peptide I am using Thermo Hypersil BDS C18 column 250 * 10 mm Column for prep column can i use it. We also have Supelco biowide discovery column.Thank you in advance.
You can probably use the C18 column as I've purified cyclic peptides on them. The solvent system used depends on your specific peptide.
The most commonly used solvent system for peptide purification is acetonitrile, water and trifluoroacetic acid. Acetonitrile and water can be easily removed by sequential evaporation in vacuo. Trifluoroacetic acid may be removed in lyophilizer. Composition of solvent to dissolve peptide before chromatography is always adapted to individual sample. For instance presence of urea usually enhances peptide solublity.
There are no any problems to purify cyclic peptides on C18 column or its analogues (for instance, phenyl). If you have hydrophobic molecules, I'd like to advise to add 2-propanol (or methanol) to MeCN (1:5).
