Hi, I'm looking for some clarification on how to make PEG-agar plates. In the protocols I've looked at, they've mentioned using filter paper or nylon mesh in between the solid, PEG-less agar and the liquid agar with PEG, and letting the PEG diffuse from the liquid to the solid media. My question is, what kind of filter paper or nylon mesh would you want? E.g. what pore size for the nylon mesh would be ideal? Any help would be greatly appreciated. Thanks!
