Currently I am working on a new protein in fission yeast. Based on the predicted function, it is involved in transcription. We found that when this gene is overexpressed, the cell growth is affected. When this gene is deleted, the cells can't grow well too. May I know what is the possible reason that causes this?

This transcription factor is probably needed for the regulation of essential genes. Therefore, cell growth is effected by the deletion.You could identify differentially expressed genes between the wildtype and the knock-out strain. These could be targets of your protein of interest.
The cell growth will be affected by every overexpressed gene due to high costs for protein biosynthesis. Additionally, your transcription factor will increase or decrease the expression of one or several other genes. Therefore, the general gene expression pattern in your cells is imbalanced. It is impossible to predict the specific effects on the cell growth until you identify the targets of this transcription factor.
