Suppose you have 15 ml falcon containing FBS. To prepare 50 ml media you require only 5 ml of FBS. Next day if you want to prepare another 50 ml media, then is it required to inactivate FBS again present in the 15 ml falcon.
People have also suggested that inactivation of serum is not necessary? How much is it true? Whether FBS should be inactivated while preparing medium for A549 cell line?
Some people suggest to make small 5 ml aliquots of fbs everytime you have to prepare media and prevent freeze-thaw cyles for multiple times.
Hi, you can deactivate the whole falcon/s and prepare your media from there. You may frozen 50ml aliquots and keep one of those in the fridge.
Ok. Firstly it is not required/ Compulsory to deactivate FBS specially in case of A549. I am also using A549 cell line. And I am using normal FBS which is of South american origin. Cells looks fine with it. So, If you have 15 mL FBS, prepare media (5 mL for 50mL media) according to your requirement.
Heat inactivation need to be done once. No inactivation is required in subsequent use.
