Good day. An immunocytochemical stain for CK20 of urothelium was perfored. Counterstain was Mayer`s hematoxylin (left figure). But after 1 day stain of the nucleouli dissapear (right figure). Why did it happen? How we can avoid it in future?
This may be an obvious question but … this is a stain that is sensitive to light, correct? How are you storing the slides? They should be stored in the dark in a cold 60 C refridgerator in my experience. Perhaps you are photobleaching them when you take the image is another option.
