I'm treating cancer cells with two different cytotoxic combination drug with low concentrations (mtt assay). I have 5 concentration (serial dilution 1:2) of each drug in triplicates. Drug A, Drug B and control group. Drug A 70% cell inhibition and Drug B 40% inhibition compared to the untreated group. I get no statistical significance between drug A and B, and a low significance between A and the control when I did t-test and Anova.
What can I do to optimize or improve the experiment or what Kind of test should be more applicable for my experiment? Because there is clear difference between viability % of the cells but non statistical wise.
-Keep in mind that drug A and B are a synergetic combination of drugs.
- Cell inhibition wasn't dependent on the dose decrement. cells reacted differently to different dose due to the presence of anti-oxidant in the combination.

- How often did you execute your experiment?
- How big is the varianze for each treament.
Sometimes - with a too big variance - the t-test wont be significant. If you give me more detail on the data you got, maybe I can help you then.
Statistical analysis will tell you there is a difference but the difference is not significant. As you observed from your data there is a difference but it is not significant.
Secondly, the inhibition data you have for the combination should be used to calculate Combination index (CI) this value will tell you the nature of combination (synergism, addictive ir antagonist)
