I am using the CellBioLabs kit to do my soft agar assay.
I am quite surprised to see in the protocol the time incubation for NIH3T3-Ras transfected cells is 8 days? It seems to me quite short.
I have done one experiment using the kit and these transfected cells as positive control, but I can't see colonies (10 days). I don't know the transfection efficacy of my transfection (no FACS, can't do immunostaining at that moment)
Have you ever experience in this kit?
Possibly to have some pictures of this positive control (in time).

Some pathogens have a different behavior regarding commercial kit chosen. Have you tried other kit to observe the pathogen's behavior?

By pathogens , what do you mean? My cells?
No I don't try another kit to see if I reproduce this issue... I was wondering if adding complete medium above my cell layer could induce this issue... I will try the next time.
I will ask the question as well.
