I am looking at studying the connection between NF-kB and JAK/STAT3 in normal and cancer cells. I am pretty certain from my research that there is a connection with IL-6. What type of tests should I propose to test this and what would some expected results be?

In addition to IL-6, other potential cytokines may include type I IFNs and IL-10. Therefore, I would look for expression of these genes in normal versus cancer cells where NF-kB is often constitutively activated. Alternatively, activate NF-kB by treatment of normal cells and follow activation of JAK/STAT1/STAT3 pathways.

So my hypothesis is that affecting levels of IL-6 will lead to decreased expression of NK-kB and STAT3.
What types of tests could I run to prove this and what other cytokines and controls would be recommended to validate my results?
