I am studying effect of DPSCs on T proliferation using PBMCs. The method I want to adopt used PHA as mitogen to activate PBMCS for two way MLR but for one way they have not clearly mentioned whether the PBMCs were also activated with PHA. Please could one also activate PBMCs with PHA in one way MLR?

There is no need to actívate T cell proliferation with mitogens to study one or two way MLR. If you will quantify T cell proliferation induced in MLR you have to be sure that there is no other stimuli that also induce proliferation. Be sure to block stimulating cells to prevent their proliferation. Good luck
