I come from the keratinocyte field, and now do murine macrophage primary culture. I was wondering why we put cold PBS-5mM EDTA and incubate it at 4°C to detach them from the Petri dishes and not 37°C. 4°C is a huge temperature shock. Does it help? Isn't it too harmful for them and their molecular expression? I asked to my colleagues, but they couldn't tell me exactly. I thank you for your answers.
