Dear Anna, I have this protocol I have been using for a long time. I have successfully knocked down some genes most of the time (around 80-90%). But I must tell you that every gene is different and the small interfering RNA @you designed makes a big difference. Some of the genes are really hard to knock down (just to let you know) but giving it a try is worth it.
Hi Anna, I think you can give it a try. shRNA molecules are processed within the cell to form siRNA which in turn knocks down gene expression. So, it might be that you can transfect cells with the protocol I have provided to you and use shRNA with it.
