21 September 2020 4 2K Report

Hello everyone,

In the paper,

Petsch et al., Nature 2012

mentions these points,

•mRNA is naturally recognized by innate pattern-recognition receptors (Effective immune response. Reducing the need for adjuvants)

•mRNA allows production of complex antigen insitu and efficient access to MHC class I antigen expressing cells (Because of genetic format. Major limitation of traditional protein based vaccines)

BUT,

the preprint article in Nature (Mulligan et al., Nature 2020), describing the Phase1/2 trials of BNT162b1 mRNA vaccine mentions,

Vaccine RNA can be modified by incorporating 1-methyl-pseudouridine which dampens innate immune sensing and increases mRNA translation in vivo

Arent these two statements contradicting? could someone please help me in understanding it.

Thanks and bes twishes,

Saif
