Hi All,

I am doing immunostaining for DNA hydroxymethylation (5hmC). I use routine immunostaining protocol. Fixation, permeabilisation, antigen retrieval, blocking, primary and secondary antibodies. When assessing slides, I see moving parts within the nuclei. Please see the videos. consider seconds a bit later to see till I fixed my hands and arms on table. I thought this was bacteria contamination. But if so they would have been all the slides? not within the nuclei only, right? Cells are fixed so are dead. No any live mechanism. What do you think what they could be? I repeated experiment 3 times, all the same. I also do DNA methylation (5meC) staining (not co-stained with 5hmC) but did not see any movement as this had.
