I haven't noticed monocyte and macrophages in the PBMCs as such. Monocytes are the circulating cells and macrophages are the tissue resident cells. when you look at freshly isolated PBMCs on glass slide you will find small circular cells, larger circular cells and larger ovoid or somewhat irregular cells. smallest circular cells are the lymphocytes and medium sized circular cells are monocytes and ovoid irregular cells may be neutrophils or other granular cells. after plating the perfect circular monocyte turns into to bigger ovoid macrophages. very rarely you find monocytes and macrophages side by side with clear demarcation. we generally believe that when it is suspension they are called monocytes after adhesion we call them macrophages.
