I am producing proteins from e. coli (BL21-DE3 strain, pET14b vector, IPTG induction).
Usually, I had enough amount of proteins (50mg of proteins from 1L LB media of 100mg/L ampicillin concentration) but recently, I had a problem with those growing conditions. (Recent my experiments gave me more cell weight - almost double amount of paste with darker color, but after the lysis I could not get anything... looks like empty vector expression)
I assumed that the depletion of ampicillin during my induction causes the problems so I increased the ampicillin concentration to 300mg/L (3 times more) and I got the nice cell paste.
Does anyone know what is the mechanism of ampicillin? (any suggestion for me - papers or books or online website)
I know it is really basic thing but I do not have background on microbiology so I need some source to learn them.
