What kind of assay is this core running for you? On a Luminex system you can run all kinds of binding assays (both protein and DNA). If you say culture sups, I would guess it is something like cytokines, concentration measurements of something your cells produced? Than I would guess they will run a dilution series of a known standard of the analyte, then measure your samples and your the standard line to calculate your concentrations. Basically, the Luminex is a multiplex ELISA, so you get multiple answers in one tiny drop of sample, reducing hands on time, and your precious sample material. And everything measured in one go, so less plate variation etc.

R.P. Achterberg - thanks, I forgot to mention. This is for protein in the supernatants. I've done ELISA once before, but it was years back...since we're paying a lot for this Luminex kit, I want to make sure I do it right.

I guess you can specify the proteins you want to let this core measure in your samples. If so, I think they will report calculated concentrations, so you don't have to worry about calculations. If they report raw data, you'd have to construct standard curves (for each analyte individually!) from the data they supply, and than use that to calculate your unknown samples,
