Hi all,

I have a few questions regarding the plasmid yields after miniprep. My vector backbone is a pET vector which is low copy number plasmid. I have been getting low plasmid miniprep yields such as 30-40 ng/uL. These concentrations were initially working out fine for Sanger sequencing with T7 forward primer but over the past few months we have not been getting any sequencing reads back.

So, my questions are:

1. What are the average plasmid yields obtained after miniprep? It would be great if you could mention conditions such as vector backbone, host cell and miniprep kit manufacturer.

2. For Sanger sequencing, what concentrations work good? I am really struggling to get proper sequencing done and I can;t rule out any factor that is affecting my results.
