my control and experimental cells, both show a strong LC3II on blots, however the experimental set shows an increase, and the LC3I band for both arises only on long exposures and that too is faint. does this mean basal autophagy rates are higher here?
i use 2% FBS during my treatments. could this (reduced serum) cause an upregulation in basal autophagy?
You mean that both LC3-I and LC3-II increased in amount?
You have to keep in mind that just because LC3-I is up-regulated does not mean the enhanced level of basal autophagy.
The important point is the ratio of LC3-II/LC3-I and further, you have to perform the experiment with CQ to stop autophagy flux and precisely evaluate the autophagy activation!

no not increased lc3 I and ii both, only increased ii form,
what I meant was I need to take longer exposures for getting the lc3 I form also, so probably lesser expression of the I form
