Is it really important to know the Splicing sites position (5 prime or 3 prime)? As we already know, one possible reason might be that we can know from which region of the gene the introns were removed. is there any other potential benefits of knowing splicing site position other than that?

said, knowing splice sites would give you some of the following information:1.) Allow you to design primers to determine splicing efficiency and also the inclusion of alternative exons.
2.) Identify effects of mutations which might disrupt splice sites to disrupt coding sequences.
3.) Allow you to investigate the pre-mRNA properties and dynamics. For example you can design probes against introns and perform nascent in-situ type experiments to determine transcription, splicing and export rates.
