Hi Rachel, thanks for the response. My understanding is that CD62L is expressed on both naive and memory T cells, and it's expression is only reduced upon T cell activation. I do not know the relative CD62L expression level between naive, activated, and memory T cells however.
I plan to remove activated and memory T cells with other markers follow by positively sort using anti-CD62L, I think this will give me a purer population of naive T cells for my adoptive transfer experiment.
It is possible. However trying to sequentially sort immune cells would take time and would have an impact on the quality of the cells downstream (esp. if planning to do functional assays). It might be better to use one of the magnetic bead isolation kits (Miltenyi, Stemcell Technologies). Negative selection is always better.
