In my opinion ..There is two methods..Either you sliced thin section of tubes and kept on the plates for incubation or second, you crushed the tubers and made dilution for the isolation process in both the conditions, sample must be surface sterilized.

After surface sterilization, plant material must be incubated in nutrient broth to check any epiphytic growth. If no growth occurs after 1 or 2 days, take out the plant material and after teasing, the inner tissues is used to inoculate on any media t o get an endophyte..
