I prepared a custom media with almost all essential components added individually (glucose, glutamine, all other amino acids, sodium pyruvate, sodium bicarbonate and HEPES), for an experiment. I do this according to a company-based formulation for the same type of medium. When checked the next day the cells seem disintegrated/non-viable, as in these pictures. I adjusted the pH to about 7.5 (pH of the DMEM:F-12 Gibco in which these cells grow well, is around 7.5-7.7). The next day pH changes to 7.39. I am guessing this shouldn't be a problem. But I am not sure what is affecting the cells. These are fallopian tube secretory epithelial cells.
I tried this twice and same thing happened.
