I have done spheroid culture with various cancer cell lines
The spheroid was formed from single cells.
The method was like the added picture (not curved bottom well, matrigel, etc)
The question is, can I do the spheroid culture with normal fibroblast(or stem cells, myofibroblasts) with the same method described in the picture?
Of course, the media composition has to change a bit.
So, the point is, I have to make a single-cell originated spheroid with cells that are not cancer cells.
Do you have any suggestions or any paper describing the method?

There are dedicated paper on homotypic "monoclonal" spheroid. You can type "CSC spheroid". Most of this kind of spheroids are generated from a single cell.

Fibroblasts in 2D culture stop growing when they become congruent. This is called contact inhibition. In 3D this effect is even stronger. Cells are lost from the spheroid resulting in 86% loss, exponentially, over a period of about a week. The remaining 14% lose clonogenicity exponentially at a rate of 90% per day (i.e., 10% surviving fraction per day of contact). Hence you will not be able to grow fibroblasts in spheroids. We have patented the Hybrid Spheroid assay in which we mix fibroblasts with tumor cells to produce Hybrid Spheroids which enable the Cancer Stem Cells to grow. See my patent for details.
