I was doing literature survey for a single & specific heamatopoietic stem cell ( HSC ) marker, that can be used in flow cytometry to detect and isolate them. Meanwhile, i have found bunch of markers , those can be used to isolate HSC but combination of these much markers to detect single HSC make the task more complicated.
Is there anyone here, who have used any single specific marker in FC to detect HSC ? Or any suggestion ?

There is no single marker that absolutely defines HSCs or any species type of stem cells , Usually we need to positive and negative marks to identify stem cells type.

there are some highly specific markers like HoxB5 (https://www.ncbi.nlm.nih.gov/pubmed/26863982) and Pdzk1ip1 (https://www.ncbi.nlm.nih.gov/pubmed/27590115). Maybe you can get a breeding pair of these mice. For humans, it's usually CD34+CD38-CD90+ lineage- CD49f+ (https://www.ncbi.nlm.nih.gov/pubmed/?term=21737740). Hope this helps.

I isolated mouse hematopoietic progenitor cells using magnetic beads. They were lineage negative and c(kit(CD117) positive cells. Look at this paper. hope it will help

It is a combination of markers (Lin- Sca-1+ cKit+ CD150+ Cd48-) and there is no single marker
