while doing MTT assay in human PBMCs with doses increasing from 25microgm/mL to 100 micro gm/mL, I have found an increasing trend in cell viability. I could not find any reference or the reason behind the proliferation.
I find the question very practical and interesting.
Several studies suggesting that curcumin and its derivatives have the capability to act as an immunomodulator. It might possible that curcumin at the particular concentration caused the lymphocyte cells to synthesize DNA more rapidly and stimulates them to enter the cell cycle, just like Concanavalin A (Con A).
Why don’t you check the cell cycle markers along with the expression of Ki67? As we cannot rely on the MTT assay, sometimes it is also because of manual errors. Western blot and RT-PCR analysis will give a clear idea.
