MTT assay demonstrated that there is no effect of a curcumin derivative at 24 h but shows good effectivity at 48 h. I think this might be due to the slow dispersion of the drug in the cell culture media (Ham's F 12 k). Please help me find a reason with reference.

I do not have a reference. However, here are some suggestions.
If you can tag your drug with a fluorophore. it will be possible to analyze the trafficking of the drug via fluorescence microscopy.
In the other hand, you can consecutively treat your cells with the drug and incubate the cells with drug treated media for for about 30-minutes 1 hour and then in different cell culture plates you can change cell cell media at different time intervals (i.e. - 1 hour, 2 hour, 3 hour etc) and check whether the effect of the drug on cells change significantly.
