Can there be any interaction (either during normal or abnormal conditions) between metabolites like that of TCA cycle (fumarate, aconitate, etc) or pyruvate, hydroxyglutarate, etc. directly with the DNA residing inside the nucleus so as to either regulate its expression level or cause structural distortion or any kind of DNA base modification?
Metabolites are of Primary and Secondary. The primary metabolites were have more DNA interactions than secondary ones. The interaction may be mutation, Carcinogenicity can occurs.. But not all the metabolites will do..
Metabolites of TCA are short carbon acids that are used mainly for energy production in the mitochondria or for building up other metabolites in the cytoplasm. However an example for physiological interaction is that between different protein and steroids that act as transcription factors and as regulators of transcription. Another form of interaction is the genotoxic interaction between electrophilic substances and DNA bases. These substances involve aromatic amines, polynuclear aromatic hydrocarbons, nitrosamines, chlorinated hydrocarbons, azo dyes and nitrosourea.
