Just asking your opinion, I already extract my sample using microwave assisted extraction. the solvent is distilled water. my target compound is mineral analysis, type of phenolic acid and also checking for total phenolic content & antioxidant activity. the solution extraction is centrifuged to get supernatant for further testing. Can I directly use the supernatant(solution form) for test without purification process (rotary evaporator/freeze dryer etc)? The instrument I use is more to spectrometer concept like AAS,ICP-OES,UV-VIS and chromo like HPLC-MS. We know that spectro is able to detect the specific element by using certain wavelength (should be no issue if not undergoing purification process). Am I right?
Correct. The HPLC is very good a purifying the sample (crap elutes at the solvent from or never elutes) and quantitating your target molecule.

In my opinion, you must go for rotary evaporation or lyophilization, because you need to prepare known dilutions (eg. mg/ml) for antioxidant activity analysis etc...
and you can perform GC-MS or NMR for identify your compounds.
If you wish to don't go for purification its okay but you must need to prepare dry sample for further applications.

thanks for your answer. may I know what do you mean with dry sample for further application? actually in sample preparation area, my sample is freeze dried and then proceed with extraction process (it become liquid), So should I freeze dried once again after extraction to become dry sample if don't go for purification? some of the people question me about how do you determine the total phenolic content if you don't remove solvent (purification) in your solution extraction even my solvent is water. Do you agree if I said no need to purify sample if our test undergoing by using spectrometer instrument such as icp-oes,aas and uv-vis?my reason is these instrument have specific wavelength to detect the specific compound, so there is no issue either our sample is purify or not..

Drying your sample is needed to conduct your tests with known concentration of sample. However compounds usually more stable in solid state more than solution.

thanks for your answer. In my research I used 1.2 gram of pitaya peel powder to extract into 50ml solvent of water. In this research I used MIcrowave assisted extraction to extract active compound from pitaya peel and then determine total phenolic content(tpc) and antioxidant. Let say my uv-vis reading for tpc is 12g/ml..so the final result is 12g/ml divide with (1.2g/50ml)x100=12/2.4=5g/ml...am I right?
