We are designing a nucleotide probe for human gene. We need to then cut off the probe from the substrate. Yet the photocleavable space is tooooo expensive. We are thinking to use an endonuclease. Is there any of the endonuclease that do not cut human sequence?
There is really nothing special about human or mammalian DNA that would make it resistant to any endonuclease. But the human genome has been sequenced, so you can check the sequence of the gene of interest to see if it has sites for the common endonucleases you might want to use.
Not likely. Maybe you could find one based on methylation patterns, but when you get down to level of ATGC, all DNA is made of the exact same molecules. Nothing special about human DNA at that fine of detail.
