I am checking the effect of inhibitor of protease which is hypothesized to degrade some proteins. I used the DMSO as a solvent. But when I add DMSO+ Bacteria there is no degradation of these protein.

Yes, there is an effect of DMSO on the bacterial growth as it is sulphoxide (containing sulphur S=O linkage) is toxic to bacteria. Hence, it is advised to use the control of the solvent.
Has DMSO been reported to show antifungal activity of its own? What would be the case in case the azoles are dissolved in DMSO for MIC determination?
Absolutely. We often do screening for antibacterial testing with compounds dissolved in DMSO, and try to stick to less than 1%. Always worked. If not freeze drying is always there to remove DMSO.
Thank you for your response. I worked with DMSO concentration less than or equal to 1.6 percent and have not seen any antibacterial activity in this range as mentioned by you.
I used DMSO with concentrated,about 10%, even about 50%. But, i found nothing perspective to antimicrobial effect of DMSO against E.coli and Stapylococcus aureus. E.coli and stapylococcus are the bacteria which have high resistance to antimicrobial,even these bactera are resistance to DMSO at every concentration.So that i used DMSO as negative control and i got zero zone of inhibition. Even i used DMSO as negative control and solvent for ether and ethanol crude extration of selected plant, i found rsistance effect of E.coli and staphylococcus aureus. In respect to my study,i refuse to agree with DMSO can gainst bacterial growth(E.coli and Staphylococcus) Thank you!

What strain of MRSA you are using? At 50% you are not seeing any inhibition. I have never heard something like this. This sound really strange. I mean really really strange.

I'm working with DMSO as solvent for some products which are used on 10 strains of Lactobacillus rhamnosus and 9 strains of Escherichia coli.
I tried those dilutions: 20%-10%-5%-2.5%-1.25%-[...]-0.3%. The strains don't react the same way, but I never saw an inhibition on the bacterial growth under 2.5% after 48h incubation. As Navid said, try to stay under 1%!
Maxime, are you seeing any inhibitory effect of DMSO on any of your strains?
Of course! After 48h, with 20% DMSO, none of my strains grew. With 10%, most of the strains where inhibited too (only one strain grew). From 5%, a good part of my strains started to grow.
I`ve used DSMO as s control for reference strains of Candida albicans, Staph aureus and E.coli, and alway had inhibition zone on agar.

Has anyone observed any effect of sublethal concentrations of DMSO on bacterial gene expression or transcriptomes? Or even the effect of DMSO on other phenotypic traits such as biofilm formation, motility, etc...?
