I have expressed this protein in E.coli and have been trying to look for an assay to measure its activity and am having difficulty finding something. this is an enzyme which converts oxidosqualene to lanosterol.
Reverse-phase, or perhaps normal-phase HPLC should be able to separate the substrates and the products. Detection could be by MS, but since both substrate and product have the same mass, separation will be essential. There should also be some far UV absorbance due to the double bonds, so UV detection may be sufficient.
