I have electroporated staphylococcus cells with a plasmid using the COL and RN6390 strains and plated onto TSA Chloramphenicol and incubated at 37degrees, however after 4 days I have no growth. How long do these cells normally take to grow?
You should probably see growth by now. Did you make control plates with the same electroporated sample plated on TSA without antibiotic?
No I didn't include any control plates. This is my first time working with staph, next time I will Include a control too.
Normally you should see growth after 1 day, sometimes it may take 2 days. If you do not see any colonies after 2 days, you will most propably not get positive clones later. If there are colonies grow after 4 days its most likely that they grow because of degradation of the AB or you get spontaniously resistant mutants, because CM is bacteriostatic and it doesnt kill the bacteria.
Staphylococcus usually takes 24 hours to grow, perhaps you should consider a control for your experiment as suggested by J. Harry Caufield
