Is there a problem to use lentivirus to infect HeLa cell line?

I had successful experience using lentivirus to infect 293T and ThP-1 cell lines.

I am recently using pEasiLv system infecting HeLa cell but never got a single positive cell. I repeated 3 times but nothing happened at all.

Does anybody have experience with HeLa cell infection? I thought it is common and should not have any problem with the infection. Is there something special to HeLa cells?

I used pEasiLv backbone for transferring DNA, pMDG for env, p8.9 for packaging.

Hi Siming,

I am currently trying to perform lentiviral transduction of THP-1 cells but I did not obtain any infected cells. How did you get it?

I tried spinfection of these cells (2000rpm 1h RT) in media supplemented with Polybrene 8ug/mL and different virus dilutions. After this spin I refresh media and made the readout of %alive cells applying Puromycin 72hrs later.
