We mainly work with primary cells and the only system that has successfully been use for KO or O/E experiments has been the use of lentiviral systems. However, our lab is abandoning the use of HEK293T lines entirely due to ethical concerns. Our PI has asked us to look into other viral packaging cell lines. After some lit search, I only come across lines derived from HEK, which our PI doesn't want to use. Could anyone please recommend other cell lines if available and share your experience with them.

I definitely do not agree with him on this and had a discussion with him about how I think this is not fair and it is creating unnecessary challenges for ongoing projects. If you have any ideas on how I can challenge his views and possibly convince him to change this decision, I would greatly appreciate it.

In HEK293T, packaging may be difficult for certain tumor suppressor genes. In each case, packaging methods using cell lines other than HEK293T are sought.
