I am culturing K562 cells In RPMI medium, they are proliferating but they are making cell clumps. Do they usually make so? If not, then is there any problem with them before going for my experiment and then how to avoid clumping?
Clumping of K562 is less than other leukemia cell lines; a low level of clumping is normal. How is their adherence to plastic? Did you check for mycoplasma contamination?
Clumping cell is not normal. Several factors may contribute to clumping. To start with what kind of culture plates/flask did you use? It needs to be uncoated plates. Further, mixing cells vigorously may reduce the extent of clumping. The extent of clumping is going to influence activation of certain cell signaling pathways. Therefore, little consideration into the goals of your cell culture, could help in achieving your goals.
yesterday K562 cell were looking good and completely attached to the surface of culture flask. But, today morning all the cells have been detached from the surface and formed several clumps. Is this normal phenomena or some problem/contamination is there ? Kindly suggest me....
