I want to study differentiation of CD4+ T cells in vitro into cytokine secreting Th1/Th2 subset. Is there a good protocol for this ? Is the CD4+ SupT1 cell line good for this study ? Thank You.
The short answer is-NO. The cell line according to ATCC has a number of genetic alterations that are likely to impact on the cell cycle and differentiation of cells. Therefore, if there is no other option for addressing the question(s) and the goal is to learn something (rather advancing the field), the cell line might be ok.
I am not familiar with this cell line. But a quick search like Divaker Choubey mentioned shows that the cells are positive for multiple T cell lineage markers (CD3+CD4+CD8+). I would not use them as a starting point for T cell polarization studies. It is fairly easy to isolate naive CD4+ T cells from donor blood and polarize then in vitro into Th1/Th2. This would also be physiologically more relevant. See the threads linked below for details.
