To do TEER analysis, I am a little concerned that whether or not I can measure it because my cells are not adherent and don't grow in monolayer form. They are suspension. However, every paper I'm looking at used TEER for monolayer cell types.
Do you think I can used this technique for the suspension cultures too?

TEER measures the resistance of an epithelial sheet to the passage of an electric current. Generally, this property is in part due to the lipid bilayer of the cells and the tight junctions between adjacent cells. To measure, you have an electrode on either side of the cell sheet. If there is media/buffer between cells, then there will be no resistance.
What cells are you trying to measure TEER on?

Thanks a lot. my cells are BY-2 cells (plant cell line). i don't know if this method is viable for them or not. it looks i need to have them out of media to measure TEER. Right?

I'm not sure about your plant cell line. I've only used this method for adherent mammalian cells. In my system, I grow the cells in transwells and allow them to reach confluence. Then, to measure TEER there has to be media or buffer inside the insert where the cells are growing and outside of the insert. However, the two environments need to be separated, or there will be no resistance at all.
Basically, you cannot measure without media/buffer. What device are you planning to use to measure TEER? I would recommend that you search for articles that have measured TEER in plant cells, and follow their method. If not available, you'd have to develop your own method.
