I am faced with an experiment that involves a large number of mice that require elispot and flow cytometry to be done on the same day., n=4 x12 groups, stimulated with 10 different peptides. will pooling of the spleens for each group be a problem, if the mice are inbred mice,. same aged?
repeat experiment 3 times to get conclusive results.
I think it's a common and accepted practice. Our group has published results with pools in the past. The drawback is that you lose the individual variations so it would be useful if you can have another correlate to show that your groups are somehow homogeneous (like individual antibodies for example)
I agree with Francisco. It really depends on the variability of your mice. For NZB/W F1 lupus mice for example this variability can be pretty high. So I would suggest to test this in advance and if the results for individual mice do not vary that much I think pooling shouldn't be a problem.
It is highly acceptable and considering your situation (several groups), it would be a feasible way to test it all. Moreover, it will help you as the assays will require a considerable number of cells.
On the other hand, in order to perform statistical analysis and also to acquire reliable results, you should consider to run independent experiments (3 repetitions).
