Is necessary to supplement the culture medium with defibrinated plasma for obtain monocytes?

I need isolate monocytes from human peripheral blood and with minimal contamination with platelets.

Is really necessary to supplement the means of monocytes with Heat-inactivated (HI) autologous human fibrin-depleted plasma?.

In some protocols do not use. Thanks for orientation.

The ultimate goal is to obtain macrophages from human peripheral mononuclear blood cells.
