I just have been doing some extractions of DNA but currently, my electrophoresis has been like this. The first well is the molecular weight marker and the eighth well is another sample I ran some time ago and is ok. I use a new buffer, new free nuclease water, I cleaned my pipettes and so, but now that I put another sample in the eighth well as control I think my DNA is degraded but I don't know.
I have to say the voltage I used is 44 because the power supply doesn't give more.
If sample 8 is the same species dna then I would say that the other samples are degraded but are high molecular weight and will be fine for pcr. Low voltage is not a problem and usually gives better results than high voltage so your running parameters are only a problem if the power supply is delivering high current and not reporting it. The degraded samples are smiling badly which could mean too much salt in the samples but the smaller size compared with 8 does suggest degradation
Diana Laura Flores Romero Unfortunatelly, the degradation will go further even at +4oC, so, very soon the sample can be unsuitable for PCR. It is better to try 2-3 different methods for DNA extraction.
Hello. I think you are puncturing the wells while loading the samples.
In gel pics seems to be well position 1 to 07 showing high concentration and impurities you go for repurification than compared to lamda dna on the basis of gel image you can better conclude dna degraded/impurites.
Lane 2-7 contains smeared DNA, you have to re-purify it properly. My suggestion to you that you should follow different procedure for DNA extraction.
Try to purify the DNA and load. Or try decreasing the amount of DNA. Use different conc of DNA n run check for the sharp band. Else sample contamination.
it does appear that there is smearing in the lanes where you loaded your samples. This can be a sign of DNA degradation. Other possible causes of smearing include overloading the gel.
To confirm whether your DNA is degraded, you could try running a gel with a ladder that contains DNA fragments of known size, and compare the band pattern to what is expected. Additionally, you could try using a different method of DNA extraction to see if the issue persists.
