Is Transwell culture of Calu-3 cell line passage dependent?
Is anyone working with Calu-3 cell line doing transepithelial resistance (TER) measurements? Do you use the cells until passage number? Is there some limit to the cells became in a barrier for the TER experiments? Until when can I work with my cells?
I usually get these cells from ATCC from around passage 22-25. I will use them up until passage 40 without a problem. I'm not really sure if I've seen any subjective data on why you shouldn't use these cells further out. From my own experience, sometimes the TEER will decrease at air liquid interface at higher passage number. Liquid/liquid interface TEER tends to remain relatively constant.
I haven't worked with Calu-3 cells. But I would like to mention that cell passage number is a conventioanl but not appropriate measure of the in vitro cell age for comparison purpose among different groups, as someone may prefer to passage cells early in growth phase while others late, resulting in discrepancies in overall passage numbers. A more accurate measure for comparison purpose should be population doubling level (PDL), which is the cumulative doubling numbers of cells calculated from all passages.
